چکیده

The mixed disulfide of lysozyme and cystine can be prepared by reaction of fully reduced lysozyme and cystine at pH 9.5. The mixed disulfide is enzymatically inactive and contains 16 residues of half-cystine per molecule. On reaction with cysteine, mercaptoethylamine, or mercaptoethanol, the mixed disulfide can be converted to active lysozyme. The initial rate of reactivation varies with pH, temperature, concentration of disulfide, and concentration of mercaptan; under the best conditions, 50% of the original activity is regenerated in 6 to 7 min. The extent of reactivation is generally between 70 and 85 % under a variety of conditions. On the basis of kinetic measurements and characterization of the lysozyme formed throughout the reactivation process, a tentative mechanism for re-formation of the correct disulfide bonds in lysozyme has been proposed. Chromatographic analysis of the reactivated enzyme shows that it contains two major components. One chromatographic component is indistinguishable from native lysozyme, but the other has about 50 to 60% of the specific activity of native lysozyme and it is not converted to native lysozyme on further reduction and oxidation.

Radiation-produced aggregation and inactivation in egg white lysozyme.

Chromatographically homogeneous egg white lysozyme has been subjected under reduced pressure to 0.67-m.e.v. y-rays. At 37% destruction of enzymic activity, three inactive aggregates and one partially active fraction have been isolated by salt precipitation and chromatographic procedures. The aggregates, upon reduction with 2-mercaptoethanol and reaction with iodoacetic acid, give derivatives wi...

The Disulfide Bonds of Egg White Lysozyme (muramidase).

The availability of the amino acid sequence of egg white lysoeyme makes it possible to identify the pairing of the 8 half-cystine residues that give rise to the four disulfide bonds present in the native protein (1,2). In establishing the positions of the disulfide bonds in insulin (3, 4) and ribonuclease (5, 6) it was shown that disulfide interchange, which occurs under certain experimental co...

Inhibition of Amyloid Fibrils Formation from Hen Egg White Lysozyme by Satureia Hortensis Extract and its Effect on Learning and Spatial Memory of Rats

Background & Aims: Alzheimer's disease is a neurodegenerative disorder characterized by the abnormal aggregation of amyloid-β plaques in the brain. Although several studies have been done for finding effective medicines in the treatment of this disease, a drug that inhibits amyloid β aggregation and ameliorates the disorder has not been approved so far. One important therapeutic approach is use...

Study the Interaction of Ni Complex of Tetradentate Schiff Base Ligand with HEN Egg White Lysozyme

AbstractInteraction of Ni complex(Salen= N, N´-ethylene bis(salicylideneimine)) with hen egg-white lysozyme (HEWL) was studied by absorption spectroscopy, competitive binding study and thermal denaturation study. The protein binding affinity of Ni complex was found to be (3.0×103M−1). The binding plot obtained from the absorption titration data gives a binding constant of 2.4 (± 0.3)×103 M...

ABSTRACT: This paper presents the results of studies concerned with the preparation of fresh food foams based on egg white and various hydrocolloids including Iranian gum tragacanth, guar and xanthan gums at different concentrations (0.002-0.040% w/w). The basic physical parameters of the obtained foams such as overrun, stability and morphology of gas bubbles were measured. Viscosity measuremen...
